Investigating Ghrelin hormone Function with microPET imaging

Ghrelin is a gut hormone which activates through it's receptor growth hormone secretagogue receptor (GHS-R). This hormone stimulates Food intake, regulates glucose metabolism, modulates immunity and inflammation, improves Cardiac performances, modulates stress and anxiety and taste sensation. Furthermore, it was reported that Ghrelin stimulates the firing rate of dopaminergic neurons in the ventral tegmental area of brain. These neurons are present various regions of the brain. The purpose of this study to map ghrelin effects on DA-signaling in the entire brain, both in cells that produce and respond to DA by using 18F-FDOPA (Fluorodopa) radiotracer in healthy rats. This pilot study will provide us the necessary information to perform more comprehensive studies in the future. This study will involve 12 healthy male rats which will be injected with the [18-F] FDOPA
radiotracer, scanned on a microPET instrument with continuous blood sampling and bodily functions will bemonitored during the procedure. The study will also give insight into the number of animals required for further experiments, reducing the number of animals needed in the future. The use of radiotracers and PET allows for refinement as the technique is minimally invasive, experiment times are relatively short (a few hours), and the animals are expected to recover normal function after the procedure.
